【摘要】:ThisguideisintendedforuseinanylaboratoryutilizingPCRorRT-PCRtoamplifyanddetectaspecificnucleicacidsequence.ThecriteriausedforevaluationoftheamplificationreactionsshouldbeadministeredbyanindividualtrainedintheuseofmolecularbiologicaltechniquesassociatedwithPCR.1.1Thisguidecoversguidelines,recommendations,basicconsiderations,criteria,andprinciplestobeemployedwhendeveloping,utilizing,orassessingPCRproceduresandspecificprotocolsfortheamplificationanddetectionofnucleicacidsequences.ThisguideisnotintendedtobeastandardprocedurewithalistofrequirementsforPCRdetectionofnucleicacids.Thisguideisintendedtoprovideinformationthatwillassisttheuserinobtainingqualityandreliabledata.1.2NucleicacidtargetsforPCRincludeDNA,aswellasRNA;RNAsequencesaresuitabletargetsforPCRfollowingreversetranscriptionoftheRNAtocomplementaryDNA(cDNA).Thistypeofamplificationtechniqueisknownasreversetranscription-PCR(RT-PCR).1.3Thisguidehasbeendevelopedforuseinanymolecularbiology/biotechnologylaboratory.Thisincludes,butisnotlimitedto,laboratoriesthatspecializeinthediagnosisofhuman,animal,plant,orbacterialdiseases.1.4ThisguideconveysthegeneralproceduralterminologyofPCRtechnologyusedforthedetectionofnucleicacids.1.5Thisguideisgeneral;itdoesnotcovertheadditionalguidancethatwouldbeneededforspecificapplications,forexample,forthePCRdetectionofnucleicacidsequencesofspecificmicroorganisms.1.6ThisguidedoesnotcoverdetailsofthevariousmethodsthatcanbeutilizedtoidentifyPCR-amplifiedDNAsequences.1.7ThisguidedoesnotcoverspecificvariationsofthebasicPCRorRT-PCRtechnology(forexample,quantitativePCR,real-timePCR,multiplexPCR,andinsituPCR),anditdoesnotcoverdetailsofinstrumentcalibration.1.8Warning-Laboratoryworkinvolvingcertainclinicalspecimensandmicroorganismscanbehazardoustopersonnel.Warning-Biosafetylevel2(orhigher)facilitiesarerecommendedforbiohazardwork().SafetyguidelinesshouldbeadheredtoinaccordancewithCLSIM29-A2andotherrecommendations().

【摘要】:TheBPIisdesignedtoyieldquantitativeinformationconcerningneutronbeamandimagesystemparametersthatcontributetofilmexposureand,thereby,affectoverallimagequality.Forpropermeasurementsoffilmexposureduetotheneutronbeamconstituents,theBPImustbefabricatedinaccordancewiththispractice.ThispracticeshallbefollowedforthefabricationofallBeamPurityIndicatorstobeusedwithMethodE545todetermineimagequalityindirectthermalneutronradiography.1.1ThispracticecoversthematerialandfabricationofaBeamPurityIndicator(BPI),whichcanbeusedtodeterminetherelativequalityofradiographicimagesproducedbydirect,thermalneutronradiographicexamination.1.2ThevaluesstatedinSIunitsareregardedtobestandard.1.3Thisstandarddoesnotpurporttoaddressthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
